Detailed requirements for candidates for the post-doc position:

1. Doctoral degree in biological, biotechnological or related sciences.

2. Scientific achievements including original scientific works.

3. Experience in managing or participating in the implementation of research projects financed from external sources and supervising students or doctoral students.

4. Experience in laboratory work, including working with cell lines, cytometric analyzes, western blots, basics of molecular biology, the ability to culture leukemia lines and conducting experiments in research on epigenetics will be an asset.

Description of tasks:

Myelodysplastic syndromes (MDS) are a variety of clonal stem cell disorders characterized by bone marrow failure and an increased risk of transformation into acute myeloid leukemia (AML). We recently discovered that combination therapy with all-trans-retinoic acid (ATRA) and an antidepressant (TCP, tranylcypromine) under laboratory conditions has a strong anti-leukemic effect. Both of these drugs are used for other medical conditions.

 

Using new genomic technologies, we plan to better understand the mechanism of action of ATRA/TCP, which will allow them to be used in the low-cost and non-cytotoxic treatment of patients with MDS and AML who desperately need more effective and safer therapies.          
The doctoral student will carry out a doctoral project under the supervision of prof. Artur Zelent and dr Łukasz Szymański as part of the NCN OPUS21 project.

 

 

The scope of work as a post-doc:

- planning and carrying out research related to the project, e.g. gene expression analysis, cell line experiments

- care for the doctoral student

- substantive involvement in the work performed

-preparation of partial reports and the final report

- statistical analysis of the obtained results

- preparation of data for the procurement of reagents, materials and small equipment for public tenders

- following the scientific literature related to the subject of the project

- preparation of publications

 

Conditions of employment:

Call for a POST-DOC position funded by the Horizon2020 BovReg - Identification of functionally active genomic features relevant to phenotypic diversity and plasticity in cattle

Requirements for the candidate:

  1. Ph. D degree or equivalent in medical, biotechnology, biology or related discipline.
  2. High motivation for scientific work.
  3. Knowledge, skills with the use of techniques in the field of gene editing and in vitro cell culture.
  4. Ability to use the following techniques: cell culture (cell line), CRISPR-mediated gene editing, PCR and qPCR
  5. Experience or knowledge with CRISPR-technique is advantageous.
  6. Publications in international journals
  7. Documented knowledge of the English language at the B2 level.

Research topic:

It is well established that epigenetic modifications are implicated in the regulation of gene expression. However, the associations between these modifications and complex phenotypes are poorly understood. The EU-funded BovReg project is interested to understand how epigenetic variation in cattle translates into commercially important phenotypes. The project brings together researchers from different parts of the world to generate functional genomics data related to robustness, health and biological efficiency in cattle. Prediction tools will provide the opportunity to select cattle populations, maintain animal welfare and support precision breeding for efficient cattle production. Using several molecular approaches, WP6 (led by dr. Hiroaki Taniguchi) has the objective to functionally validate key candidate regulatory sequences and variants associated with disease resistance, biological efficiency and robustness, identified in other work packages.

Task 1: Selected candidate regulatory regions (promoters, enhancers, silencers, etc.) and genetic variants (identified in other work packages) will be functionally characterized in different bovine cell lines using CRISPR/Cas9 and other molecular techniques. After generation of the desired mutations/deletions, functional assays (qPCR and cell growth assay) will be performed.

 

Task 2: To measure interaction of two DNA regions in a high throughput manner, Hi-C-seq experiment was already performed for several cell lines by other work package. In our package, many of these interactions will be validated using 3C assay (PCR-based analysis).

 

Responsibilities and benefits in the project:

Research topic:

Parkinson’s disease is the second most common neurodegenerative disorder, affecting roughly 6,1 million people worldwide and one of the leading causes of disability. Symptoms include slowness of movement, tremor, and rigidity as well as depression and cognitive decline. While treatment options exist, they lose effectiveness over time and produce a plethora of side effects

NFE2L1 gene encodes a factor responsible for a variety of cellular functions among others the clearance of badly formed and unnecessary proteins. However, we lack direct evidence of the NFE2L1 gene’s involvement in the development of Parkinson’s disease. 

In this project, we wish to explore this possible involvement and investigate the responsible mechanism. We will deactivate the gene specifically in the dopamine secreting neurons and we will observe the effects on mice behavior. The appearance of Parkinson-related symptoms will confirm that NFE2L1’s dysfunction could be the cause of Parkinson’s disease.

We will also perform these types of analyses on an in vitro model of neuronal development. Cells will be specially treated to cause their differentiation into neurons. The cells in which we have deactivated NFE2L1 will be compared to intact cells in the number of produced neurons, their growth, and other parameters.

Task 1: Generating an NFE2L1 dopaminergic neuron-specific knock-out mice

Task 2: General observation of mice behaviour and health indicators. Testing for Parkinson’s disease-like phenotype – motor symptoms, Testing for Parkinson’s disease-like phenotype – nonmotor symptoms

Research topic: The importance of social relationships for pigs’ behavioural expression, health and fitness

 

Description of tasks:

The project involves collecting data, mainly through behavioural observations, of farm pigs; statistically analysing the data; and preparing manuscripts for publication. The data collection largely takes place in Edinburgh, UK, in collaboration with a team of researchers at Scotland’s Rural College (SRUC). The final part of the project takes place in Poland and involves behavioural observations on pigs. As part of the doctoral programme, there are several compulsory PhD training courses to enhance the candidate’s scientific skills. 

 

Responsibilities in the project:

The candidate will be responsible for the collection and organization of data, and for analysing and communicating the results of the project to the wider scientific community (through scientific publications and conference presentations).

Research topic: Preventive effect of oral supplementation with Sucrosomial® Iron in iron deficiency anemia of prematurity: studies on the animal model of preterm piglets

 

Description of tasks:

Collection, portioning and conservation of biological samples, performing biochemical iron analyzes (measurement of iron parameters in the blood plasma, iron content in tissue), RNA isolation, preparation of extracts from tissues, performing molecular analyzes (Real Time PCR, Western blot, immunofluorescence analyzes) first - under supervision, next - autonomously , preliminary data analysis, preparing manuscript drafts.

 

Responsibilities in the project:

We have in mind that the student is a "fresh" member of a team who has yet to acquire knowledge related to iron metabolism but also to molecular techniques. Therefore he/she is expected to have basic experience in the use of molecular biology techniques, statistical methods, teamwork skills, good communication skills in written and oral English.

The Ph.D student will work in a highly multidisciplinary environment. The student's project will be focused mainly on the construction of mouse inter-strain chimeras, and on their use to investigate how the placenta may affect fetal brain development and postnatal behavior.

Dyscyplina: zootechnika i rybactwo

Temat badawczy: Zjawiska genomiczne, proteomiczne i epigenomicznie w parenchymie zdrowych ćwiartek wymienia krów mlecznych sąsiadujących z ćwiartkami zakażonymi gronkowcami koagulazo-dodatnimi lub ujemnymi

 

Opis zadań:

Projekt dotyczy wpływu zakażenia tkanki wydzielniczej gruczołu sutkowego krów mlecznych (parenchymy) gronkowcami koagulazo-ujemnymi i –dodatnimi jednej/dwóch ćwiartek wymienia na poziom ekspresji genów, stopień metylacji wybranych genów i transkryptom ncRNA w tkance wydzielniczej i komórkach somatycznych w mleku w niezakażonej, sąsiadującej ćwiartce wymienia. Zakładamy, że patofizjologiczne procesy zachodzące w zakażonych tkankach wpływają na procesy fizjologiczne w niezakażonej ćwiartce wymienia (regulacja transkrypcji czy translacji) poprzez zjawiska epigenetyczne. Analiza poziomu metylacji wybranych genów oraz profil ekspresji niekodujących RNA (ncRNA) pozwoli zidentyfikować wpływ zakażeń bakteryjnych na regulacje epigenetyczne i zmiany w ekspresji genów układu immunologicznego (mRNA i białko). Materiał do badań będzie stanowić tkanka wydzielnicza wymienia otrzymane z osobnych ćwiartek wymienia – niezakażonych i zakażonych gronkowcami koagulazo-dodatnimi i ujemnymi. Grupę kontrolną będą stanowić próby pobrane z ćwiartek całych zdrowych wymion. Określony zostanie również związek poziomu metylacji poszczególnych genów i wzorów ekspresji mikroRNA z ekspresją genów na poziomie mRNA i białka. Niniejsze badania będą kontynuacją i uzupełnieniem wcześniej przeprowadzonych analiz profilu transkryptomicznego.

Research topic: the functional genomic studies of resveratrol, curcumin, and similar antioxidant phytochemicals

 

Description of tasks:

Resveratrol, curcumin, and related antioxidant phytochemicals with phenolic groups have preventive and therapeutic effects with respect to cancer. These effects can be studied using the ultra-modern tools of transcriptomics, as the phytochemicals induce global alterations in gene expression profiles. In particular, resveratrol and curcumin can induce transcription of genes related to cell cycle arrest and apoptosis in cancer cells cultured in vitro. Dr. Huminiecki the principal investigator (PI) on the project will start with a computational re-analysis of public transcriptomics datasets available for curcumin, resveratrol, and similar phytochemicals. Next, the PI and the PhD student will together conduct experiments to generate new transcriptomics datasets, from either normal or cancer cells treated with the phytochemicals. Our experiments and datasets will focus on dose escalation, time-courses, splicing, and alternative transcriptional start sites, as well as non-coding RNAs, which is a novel approach. We will also compare responses in p53 wild type versus p53-mutated cell lines to differentiate between different mechanisms of apoptosis. The studentship is funded by a grant from the prestigious Polish National Science Center —NCN, which guarantees a safe project, good education, and enjoyable working environment. (Moreover, the PhD position will be advertised and filled according to all professional rules, the standards of professional conduct, and legal regulations of the hosting Institute and NCN.)

 

 

Responsibilities in the project:

The PhD student will work and study in the area of functional genomics. In particular, the student will conduct experimental tasks associated with in vitro cell culture and transcriptomics. These will include routine tissue culture, the isolation of RNA, the set up of qPCR, as well as microarray and RNA-seq screens. The student will communicate with commercial providers of cells, reagents, and NGS services. The student will also undertake postgraduate coursework in statistics and bioinformatics, and go through routine safety introductions and PhD courses at the Institute. The student will acquire a set of skills that is transferable and highly sought after internationally: guaranteeing good career prospects after the completion of the course, either in academia or in industry.

Scholarship holders will be selected by a committee appointed by the Director of the Institute of Genetics and Animal Biotechnology of the Polish Academy of Sciences will take place in accordance with par. 14 of the Regulations for the granting of scientific scholarships in research projects financed from the National Science Centre of March 14, 2019 and with point 2.1.3 Remuneration and scholarships for students and doctoral students of Annex 2 to the Regulations on awarding funds for the implementation of tasks financed by the National Science Center in the field of research projects.

The Institute of Genetics and Animal Biotechnology of the Polish Academy of Sciences has the HR Excellence in Research logo awarded by the European Commission to institutions which implement rules of the "European Charter for Researchers" and the "Code of Conduct for the Recruitment of Researchers".

finansowane w ramach projektu pt. mEATquality - Linking extensive husbandry practices to the intrinsic quality of pork and broiler meat.” nr H2020-FNR-2020-2

Koordynatorem projektu jest Stichting Wageningen Research (Holandia), a uczestnikami projektu jest 16 naukowych i przemysłowych partnerów międzynarodowych (2021-2025).

Nazwa stanowiska: Doktorant

Wymagania stawiane kandydatowi:

  1. Tytuł zawodowy magistra/magistra inżyniera lub równoznaczny uzyskany na kierunku nauk o zwierzętach, zootechnika, weterynaria lub pokrewne(lub uzyskanie takiego tytułu do 30 września).
  2. Wysoka motywacja do pracy naukowej.
  3. Mile widziane doświadczenie naukowe z zakresu zagadnień związanych z hodowlą i chowem drobiu mięsnego oraz umiejętność praktycznego obchodzenia się z ptakami
  4. Wiedza, umiejętności z wykorzystaniem technik z zakresu zachowania się zwierząt i etologii
  5. Umiejętność obsługi programu SAS
  6. Bardzo dobra znajomość statystyki
  7. Udokumentowaną znajomość języka angielskiego na poziomie B2.
  8. Precyzja, sumienność i dobre zdolności manualne.
  9. Ogólne kompetencje – dyspozycyjność, prawo jazdy kat. B, bardzo dobra organizacja pracy, w tym umiejętność samodzielnej pracy, jak też pracy w interdyscyplinarnym zespole naukowym, kreatywne myślenie.

Temat badawczy: Systemy produkcji, a dobrostan i jakość mięsa kurcząt brojlerów

 

Opis zadań:

Stypendium doktoranckie w ramach prestiżowego międzynarodowego projektu mEATquality finansowanego przez Unię Europejską daje doktorantowi szerokie możliwości wyjazdów konferencyjnych, staży naukowych oraz pozwala na zbudowanie sieci kontaktów w najlepszy jednostkach badawczych w Europie.

 

Zakres obowiązków w projekcie:

- przygotowanie i przeprowadzenie eksperymentów

- zbiór danych: zachowania, zdrowia oraz produkcyjności ptaków

- analiza danych oraz prezentacja wyników w formie publikacji (co najmniej 4) oraz doniesień konferencyjnych

-obrona doktoratu
